The opening Guinness clash, on the weekend of 3 September, is away at Worcester's Sixways ground, and could see Gloucester and its new signings Mike Tindall, Peter Richards, Ludovic Mercier and Patrice Collazo, come up against both Andy Gomarsall and Chris Fortey, who have moved up the M5 this summer.
Gloucester's first home game is at home to Sale Sharks on Saturday 10 September, which is followed by a visit to newly promoted Bristol Shoguns (17/18 September).
Northampton Saints are the next visitors to Kingsholm on Saturday 24 September, before Gloucester travel to Bath on 15 October.
The fixture list currently contains five allocated slots for knockout cup rounds (the first two being 1 Oct and 8 Oct). Further details will be added once the format of the cup is agreed.
Other key dates in the new fixture list include Wasps away on Christmas Eve, and Leeds at home on New Year's Eve.
The final run-in of home games after January sees Gloucester host some of its biggest games at Kingsholm. Leicester (11 Feb), Bath (25 Feb), Bristol (25 March), Worcester (15 April) and Wasps (6 May - last game of regular season) all visit the West Country in 2006.
Elsewhere on the opening round of the 2005/6 GUINNESS Premiership, current champions London Wasps begin the defence of their title against London rivals Saracens at Twickenham in the second London Double Header.
Both teams finished their campaigns at Twickenham last season, London Wasps beating Leicester Tigers to win the Premiership and Saracens defeating Gloucester to win the Wildcard Final. They will be joined in the Twickenham show-piece by London Irish and Leeds Tykes.
The inaugural London Double Header was played in front of 51,000 supporters at Twickenham. With the relegation of NEC Harlequins, Leeds Tykes have grasped the opportunity to begin the season at Twickenham and will be aiming to replicate the success of their last visit to rugby's headquarters when they beat Bath 20-12 to win the Powergen Cup.
Gloucester's clash away at Worcester is not the only West Country derby on the opening day of the season. Newly promoted Bristol Shoguns kick off with a home match against Bath.
The first round fixtures also offer an East Midlands derby as Leicester Tigers meet Northampton Saints at Welford Road. To complete the set, European Shield winners Sale Sharks play Newcastle Falcons at Edgeley Park.
Last season's Premiership was the closest and best attended on record. In nearly half the 132 matches last season, the winning margin was seven points or less.
The last round of the season saw three teams competing for the right to meet London Wasps in the Zurich Premiership Semi-Final (Bath, Sale Sharks and Saracens) and five teams battling to avoid relegation (Leeds Tykes, London Irish, NEC Harlequins, Northampton Saints and Worcester Warriors).
Attendances rose by 15% on last year, with the average per round attendance reaching nearly 10,000 (9,978). 57 of the 132 matches were sell-outs.
Six of Gloucester's 11 home Premiership fixtures last season were sell-outs, and the average attendance was 12,427. The six 13,000 sell-outs contrasted with a lowest crowd of 11,265 against London Irish.
The season will climax with the GUINNESS Premiership Final on 27 May 2006.

The Kingsholm club has yet to confirm its pre-season schedule, apart from its involvement in the annual Middlesex Sevens tournament on 13 August.
