The two respective leaders of the Guinness A League sections met as part of Leicester's centenary celebrations and the Tigers claimed a 32-24 victory.
But a number of Gloucester's rising stars came through the examination - Jon Goodridge, who scored a try, Anthony Allen, Mark Cornwell and Terry Sigley all played major roles in the contest.
Gloucester's tries came through Goodridge and Ali James, Ryan Lamb converted one and scored the rest of the points through penalties.
"Leicester put out a very experienced side and at United level, they are probably the best team in the country," said coach Mark Hewitt.
"We found ourselves 10 points down pretty quickly after Leicester really turned it on, but we battled back really well and enjoyed the lions share of possesion until half-time.
"It was a massive challenge and we produced a really good performance. We hope to repay the favour by having Leicester down to us in the New Year."
