Although the full Barbarians line-up has not been confirmed, Vainikolo, Balshaw and Collazo will have the opportunity of one last appearance of the season at Kingsholm.
"Everyone in rugby knows what the Barbarians stand for and the traditions they hold," said Vainikolo. "To have the chance to represent them - at Kingsholm - is a fantastic honour and something I am really looking forward to.
"The Gloucester crowd is fantastic and hopefully we can give them one last memory of the season to take away with them. Personally, I could not ask for a much better way to end the season and I hope we get some great support."
Former Gloucester scrum-half Andy Gomarsall is also likely to feature alongside stars like Regan King and Felipe Contepomi.
"We are hugely fortunate to be able to stage a game of this magnitude at Kingsholm and to have some Gloucester representation in the Barbarians is wonderful," said Gloucester's managing director Ken Nottage. "It illustrates that Gloucester Rugby and Kingsholm are growing all the time and to attract an event of this size is fantastic for us.
"Our supporters have been tremendous all season and hopefully they will see this as a fitting way to end the season here."
